Integration of biological logic-based techniques for functional expression of recombinant proteins in-vivo

The need for a systematically integrated approach to improve functional expression of recombinant proteins in-vitro and/or in-vivo is still proliferating and thus continuously gained much attentions. Therefore, the related researches focused on the development and integration of novel logics, parts and modules for functional expression of POI (protein-of-interest) is steadily increasing. In this context, the elucidation of underlined mechanism for the control of gene expression at the level of transcription and translation is prerequisite for the systematic design of expression systems including recombinant vectors and expression hosts. As generally well known, cell innately controls gene expression using a set of various cis- and trans-acting elements, and the interactions between them were logically programed to modulate gene expression in-vivo. A lot of attempts, therefore, have been made to understand the logic of the genetically installed program on genome for regulating gene expression and have made great advances in related techniques. Here we briefly review and discuss the logic or mechanism of gene expression recently elucidated, and demonstrate that an integrated logic-based technology for overexpression of the target protein has great potential for bacterial anticancer therapy in-vivo. We also discuss the point that this tool was useful for the functional expression of difficult to express proteins in-vivo.
